NEW DICLOFENAC DRUG DERIVATIVES AND THEIR METAL COMPLEXES: PHYSIO-CHEMICAL CHARACTERIZATION, INSILCO AND INVITRO STUDIES |

Abstract |
The Schiff base 2-(2-((2,6-dichlorophenyl)amino)phenyl)-N'-(2-hydroxybenzylidene) acetohydrazide was prepared by condensation of the diclofenac hydrazide with salicylaldehyde and characterized by FTIR, uv-vis, 1HNMR and mass spectra. copper (II), nickel (II), cobalt (II), Gd (III), La (III) and Ag (I) complexes were synthesized by the reaction of the metal nitrates or chlorides, with the prepared Schiff base. The structures of the formed complexes were discussed according to the thermal, elemental, FTIR, uv-vis, magnetic and conductivity measurements. The obtained conductivity revealed the non-electrolytic behavior of the obtained complexes except Gd (III) complex. The magnetic susceptibility and uv-vis spectra indicated the octahedral structure of the complexes under consideration. In vitro anti-inflammatory activity of the synthesized Schiff base and their complexes evaluated by: Enzyme-linked immunosorbent assay (ELISA). Antitumor Activity Evaluation (MTT – Cytotoxicity assay protocol) of the ligand and copper (II), Gd (III) complexes was tested against MCF-7 and HepG-2 cell cancer. Estimation of physicochemical properties, pharmacokinetics and drug-likeness in silico for prepared compounds, which was performed using SwissADME web tool. The interaction of the synthesized compounds with the COX-2 (PDB code: 5IKT (Homo sapiens)) was tested and evaluated using the docking operation with aid of some computational chemistry software. The obtained results go well with the experimental work. |
