TAILORED SOME NEW CORTISONE DERIVATIVES AND EXPLORE THEIR PROMISING CHEMICAL AND BIOLOGICAL APPLICATION PROSPECTS

Abstract
New Schiff base Hydrocortisone- phenylene diamine was synthesized long with metallo- derivatives of Copper (II), Nickel (II), Magnesium (II) and Vanadium (IV) was obtained in a pure form as complexes. The compounds acquired were described by elemental evaluation, 1HNMR, FT-IR, Mass spectra, thermal analysis methods (TGA, DTG, DTA), magnetic susceptibility, ICP, and conductivity. The obtained data indicated the formation of the target complexes: [Cu(HPD)2Cl2] 4H2O, [Ni(HPD)2Cl2(H2O)2] 5H2O, [Mg(HPD)2SO4] 7H2O, [V(HPD)2SO4(H2O)2] 7H2O. In vivo and invitro studies were applied to study the impact of the synthesized compounds as an anti-inflammatory drug. The in vitro COX- 2 isozyme inhibition assays determined the ability of the target compounds to inhibit human recombinant COX- 2 using an enzyme immunoassay (EIA) and enzyme- linked immunosorbent assay (ELISA). According to the results of IC50 of HPD, Cu-HPD, Ni-HPD, Mg-HPD and VO-HPD compounds is 1.29, 1.60, 1.71, 1.40 and 1.60 pg/ml respectively, while the IC50 of hydrocortisone is 1.72 pg/ml, in vivo study applied 3 different tests; hot plate test, acetic acid test and formalin test. All studies showed a significant high effect compared with the row material Hydrocortisone.
